Caractérisation du comportement chimique du complexe otosac-otolithe et réaction à des perturbations induites. ArchiMer
Edeyer, Anaick.
Teleost fish have the capacity to record in their otoliths, calcareous concretions located in the inner ear, the principal events (environmental and physiological) which punctuate their life. Otolith is made of CaC03 deposits on a protein matrix according to a circadian rhythm. Due to the quasi irreversibility of this process, otoliths can be compared to biological archives largely used to apprehend the fish ecology. The otolith bathes in the endolymph, a medium in which all the precursors of otolith formation forward. The endolymph chemical analysis revealed a heterogeneity of its principal components (proteins, total C02 and total Ca) which origin lies in the non uniform spatial distribution of the ionocytes in the saccular epithelium. Thus, in...

Dietary lipid level, hepatic lipogenesis and flesh quality in turbot (Psetta maxima) ArchiMer
Regost, Christelle; Arzel, Jacqueline; Cardinal, Mireille; Robin, Jean; Laroche, M; Kaushik, Sadasivam.
A study was undertaken with marketable size turbot to evaluate the effects of dietary fat levels on chemical composition. lipogenesis and flesh quality, Four experimental diets containing graded levels of fish oil in order to obtain 10%, 15%, 20% and 25% of crude fat were fed to triplicate groups of turbot (initial body weight of 660 g) for 12 weeks in full strength seawater at temperature of 17 degreesC. Nutrient digestibility was not influenced by dietary fat levels. The best growth performance was observed in fish fed 10% and 15% dietary fat. High dietary lipid levels led to higher fat deposition in whole fish, although lipid level in muscle remained low (1.1% in dorsal muscle and 1.7% in ventral muscle irrespective of diet), Significant subcutaneous...

Effect of temperature, volume of ova batches, and addition of a diluent, an antibiotic, oxygen and a protein inhibitor on short-term storage capacities of turbot, Psetta maxima, ova ArchiMer
Suquet, Marc; Chereguini, Olvido; Omnes, Marie-helene; Rasines, Inmaculada; Normant, Yvon; Souto, Isabel; Quemener, Loic.
The effect of different parameters on shea-term storage capacity of turbot ova was assessed over a 45-h period after ova collection for fertilization rates and over a 9-h period after ova collection for hatching rates. Increasing the volume of ova sampling from 0.5 to 2.5 mt, as well as adding an antibiotic-antimicotic solution or oxygen did not significantly change the storage capacity of ova. Regarding the hatching rates, a higher storage ability was recorded at 8 and 13 degrees C, compared to 3 degrees C. The mean composition of the ovarian fluid was determined (n = 57 spawns). Use of a diluent mimicking the ovarian fluid significantly decreased the storage ability as assessed by the fertilization rates but did not modify the hatching rates. Diluting...

Long-term effects of the cryopreservation of turbot (Psetta maxima) spermatozoa ArchiMer
Suquet, Marc; Dreanno, Catherine; Petton, Bruno; Normant, Yvon; Omnes, Marie-helene; Billard, Roland.
The survival of turbot eggs and the rearing capacities of larvae stemmed from artificial fertilization practices using frozen-thawed spermatozoa were evaluated. Furthermore, the viability of sperm samples stored during a 9 month period in liquid nitrogen was assessed. No significant difference in the fertilization rate, hatching rate, survival and wet weight of 10-day old larvae were observed using fresh or frozen-thawed spermatozoa. The motility recorded at 10 s and 60 s post-activation and the fertilization capacity of frozen-thawed spermatozoa were not significantly decreased during a 9 month storage period in liquid nitrogen. These results confirm the high quality of the turbot spermatozoa stemmed from the cryopreservation process, allowing their use...

Total replacement of fish oil by soybean or linseed oil with a return to fish oil in turbot (Psetta maxima) - 1. Growth performance, flesh fatty acid profile, and lipid metabolism ArchiMer
Regost, Christelle; Arzel, Jacqueline; Robin, Jean; Rosenlund, Grethe; Kaushik, Sadasivam.
The aim of the study was to investigate the replacement of fish oil by vegetable oils and the effects of a washout with a return to fish oil on growth performances and lipid metabolism. three experimental fish meal-based isonitrogenous (crude protein content: 57.5%) and isolipidic (crude lipid content: 16.5%) diets were formulated containing either 9% of added fish oil (170), soybean oil (SO), or linseed oil (LO). Each diet was distributed to triplicate groups of 25 marketable size turbot (initial body weight of 579 g) grown in seawater at water temperature of 17 T. Fish were fed once a day to visual satiety. At the end of the growth trial, which lasted 13 weeks, all groups of turbot were fed with FO diet for 8 weeks. The growth of turbot was high, but the...

Total replacement of fish oil by soybean or linseed oil with a return to fish oil in Turbot (Psetta maxima) 2. Flesh quality properties ArchiMer
Regost, Christelle; Arzel, Jacqueline; Cardinal, Mireille; Rosenlund, Grethe; Kaushik, Sadasivam.
The aim of this study was to evaluate (1) the effects of replacement of fish oil by vegetable oils on flesh quality and (2) the effects of a washout with a return to fish oil on flesh quality of turbot. In a first period of 3 months, three isonitrogenous and isolipidic diets containing 9% of added marine fish oil (170), soybean oil (SO) or linseed oil (LO) were fed to triplicate groups of 25 marketable size turbot (initial body weight: 579 +/- 1 g) grown in sea water at the temperature of 17 degreesC. At the end of the first period, all groups of turbot were fed with the diet containing fish oil (diet FO) for a further period of 2 months. The gutted and fillet yields were not affected by the incorporation of vegetable oils. However, soybean or linseed oils...

Toxicity of ammonia to turbot juveniles: I. effects oil survival, growth and food utilisation ArchiMer
Person-le Ruyet, Jeannine; Delbard, Christine; Chartois, Herve; Le Delliou, Herve.
Long-term effects of constant exogenous ammonia concentrations were investigated in two different batches of turbot juveniles (53 and 73 g) under controlled environmental and feeding conditions. Over the 84-day experiments, ambient un-ionized ammonia (UIA-N, NH3) concentrations were steady (coefficient of variation, 12-16 %) and water pH range was 7.88-7.99. Survival was maximum up to 0.33 mg.l(-1) UIA-N and at 0.73, 50 % mortalities were observed on day 52 (73 g) or day 77 (53 g). No-growth concentration was 0.73 mg.l(-1) UIA-N in the two groups and the 84-day lowest-observable-effect concentration range 0.14-0.33 mg.l(-1). In the most tolerant group (53 g), the 84-day no-observable-effect concentration was 0.14 mg.l(-1) UIA-N. The estimated 28-day...
